Introduction to the solution to common problems in centrifuges

A centrifuge is an instrument often used in biological experiments. It uses a high-speed rotation of a rotating shaft to generate centrifugal force to separate substances of different densities and sizes. It can achieve the purpose of initial separation and purification and is an indispensable instrument in the laboratory.

According to the speed, it can be divided into three types: low speed (<8000rpm), high speed (8000-20000), and overspeed (>50000). According to the size and placement position, it can be divided into a desktop centrifuge and a floor-standing centrifuge. The desktop centrifuge is relatively small in size, often a medium- and low-speed centrifuge. It is commonly used in general molecular biology experiments. Floor-standing centrifuges are large in volume, often with temperature-lowering devices, mostly high-speed centrifuges, which are often used for preparation and preliminary separation. , large-scale centrifugation, and centrifugation for some required temperatures.

Proper use, prolonged service life, and minimized damage can reduce our experimental progress. We may have the following doubts.

Question 1: What should I pay attention to when placing the centrifuge?

A: The centrifuge is usually placed in a dry place away from direct sunlight. Centrifuge heat dissipation is relatively large, do not pile up debris around, surrounded by walls, baffles and other items that are not airtight, poor heat dissipation, at least 10cm. At the same time, the centrifuge should be placed in a room as close as possible. Never put organic reagents or flammable materials around. As shown in Figure 1 above, the damage of the centrifuge and the resulting air waves cause the medicine cabinet to oscillate. Fortunately, the organic reagents Did not spill, otherwise the consequences will be more serious.

Question 2: What is the most important thing when using the centrifuge, what is the most critical step?

A: After the centrifuge starts working, it is similar to a pistol. It is similar to a bomb. Therefore, in the event of an accident, it is quite dangerous. The high-speed or ultra-high-speed rotating rotor flies out and can almost penetrate everything. So nothing is the most important and crucial.

Question 3: What should we pay attention to as a user?

A: As a user, we should use a special centrifuge tube for the liquid containing organic reagents according to the centrifuge tube required for the experiment. Select the appropriate rotor, the speed is absolutely prohibited to exceed the maximum speed indicated by the rotor. The accident was caused by a mistake in the use of the rotor. The correct use of the centrifuge tube and rotor is critical, especially for the symmetrical placement and weight trimming of the centrifuge tube. At the same time, attention should be paid to the maximum weight of the centrifuge tube and the rotor. The liquid can hold up to 3/ of the full volume of the centrifuge tube. 4. Many accidents are caused by improper use of the rotor and the centrifuge tube.

Question 4: What maintenance should the centrifuge pay attention to, which can extend the life?

A: After each use, the centrifuge cover should be opened to allow the heat or moisture to evaporate naturally. If you have used low temperature centrifugation before, there may be ice. If the ice melts, wipe it with dry cotton gauze. Then cover the cover when there is no obvious water vapor. If the centrifuge rotor can be replaced, each rotor should be removed in time after use, cleaned with a clean, dry medical gauze, and placed under the buckle. Do not use a sharp scraper. The aluminum rotor should be cleaned frequently, and the centrifuge should not be maintained and repaired frequently. The operator should cut off the power when leaving. For the first time users, please consult the personnel who have used it before or refer to the instruction manual. Do not use it blindly.

Question 5: What are the common accidents in centrifuges?

Answer: Because the frequency of use of the centrifuge is high, the machine has damage and the frequency of the accident is high. The main reason is caused by improper operation of the experimenter. Common problems are: the cover cannot be opened, the centrifuge tube cannot be taken out, and the centrifuge button is pressed. Not working; more serious problems are caused by uneven rotation of the shaft, motor burnout, horizontal buckets, and more serious accidents as shown in the above figure, and even wounding, and so on.

Question 6: What should we pay attention to when doing specific operations?

A: When we use it, we must pay attention to it from all aspects. After selecting the rotor and setting the parameters, it is necessary to observe the centrifuge for a while, and then reach the maximum speed and run stably before leaving. If you hear abnormal sounds or smell any smell during operation, you should immediately take the brakes, press the “Stop” button, and turn off the power if necessary. The centrifuge tubes must be placed symmetrically, and the corresponding centrifuge tubes should be as equal as possible. It is absolutely forbidden to open the centrifuge cover during the operation of the instrument! It is also necessary to develop good registration habits for all personnel in the laboratory. The first one can know who used it before, the condition of the instrument before use; the second can know the number of times the centrifuge is used. To know if it needs to be repaired or replaced.

Although with the development of intelligence and automation, the common problems of the centrifuge, such as the actual use of the rotor and the input rotor number is wrong, or the maximum centrifugal force input does not match the maximum centrifugal force of the rotor design itself, the symmetrically placed centrifuge tube When the weight difference exceeds the limit of the centrifuge, the centrifuge automatically stops working. But don't worry about it, let the centrifuge help us check our mistakes. First, it is a loss for the centrifuge and the rotor, and it wastes time. If the sample is sensitive to temperature, the sample is not Well, secondly, if the centrifuge does not have a self-test error, if the tragedy is caused, the consequences will be disastrous. In short, centrifuges, as a laboratory-used, seemingly simple instrument, still have a lot of knowledge, we need our common care and maintenance, in order to extend its life and more stable service for our experiments.
