D-type cyclins (D1, D2, and D3) and their associated cyclin-dependent kinases (CDK4 and CDK6) are part of the core cell cycle machinery that drives cell proliferation. Inhibitors against CDK4 and CDK6 are currently being tested in a number of clinical trials and exhibit desirable therapeutic efficacy in several cancer types of patients. For example, in March of this year, Novartis's Kisqali (ribociclib, LEE011) received FDA approval as a first-line treatment for HR+/HER2-metastatic breast cancer, in combination with any aromatase inhibitor for the treatment of breast cancer.

A team led by Dr. Piotr Sicinski of the Dana-Farber Cancer Institute tested the CDK4/6 inhibitor Kisqali (ribociclib) in a melanoma xenograft model. They found that in xenografts from 33 melanoma patients, 3 patients showed long-term regression, and all 3 tumor cells were derived from patients with high levels of cyclin D3 and CDK6 xenografts. In contrast, cyclin D3 and CDK6, which are non-regressing tumors, have lower pre-implantation levels.
